Screening of Chinese herbal medicine for inducing expression of β-defensin-2 in gingival epithelium

[Abstract] Objective To screen out the Chinese herbal medicine which can induce the expression of β-defensin-2 in rat gingival epithelium. Methods 90 rats in our hospital from February 2019 to April 2020 were selected as the research objects. Ninety healthy adult Wistar rats were randomly divided into three groups: the inherent expression group (10 rats), the negative control group (10 rats) and the positive control group (10 rats). The experimental group were given drugs respectively (the astragalus membranaceus group, the Radix Isatidis group, the salvia miltiorrhiza group, the radix bupleuri group, the herba houttuyniae group, and the andrographolide lactone group, with 10 rats in each group, a total of 60 rats). After 7 days of continuous administration, the rats were sacrificed, and the gingival epithelium of the rats was cut. The expression of β-defensin-2 protein in the gingival epithelium of the rats was detected by the immunohistochemical method. Results The expressions of β-defensin-2 protein in the gingival epithelium of rats in the astragalus membranaceus group and the radix isatidis group were significantly higher than that in the inherent expression group, and the differences were statistically significant(P<0.05), but no statistically significant differences were observed compared with the positive control group(P>0.05). Conclusion Astragalus membranaceus and radix isatidis can induce the increase of β-defensin-2 expression in the gingival epithelium of rats, suggesting that astragalus membranaceus and radix isatidis can regulate the innate immunity of oral mucosa of rats.

現(xiàn)今因抗生素濫用而引起的病原微生物耐藥、過敏反應(yīng)、正常菌群失調(diào)等問題日趨嚴(yán)重,研究和開發(fā)內(nèi)源性肽類抗生素已備受醫(yī)學(xué)界關(guān)注。防御素是現(xiàn)今研究中最受關(guān)注的內(nèi)源性肽類抗生素分子,具有抗細(xì)菌、抗病毒、抗真菌、抗腫瘤等多種生物學(xué)活性,并且尚未發(fā)現(xiàn)有耐藥菌的產(chǎn)生[1-2]。哺乳動(dòng)物類防御素按其結(jié)構(gòu)不同可分為α-防御素、β-防御素和θ-防御素三種。在哺乳動(dòng)物口腔中,β-防御素主要分布于上皮細(xì)胞內(nèi),是宿主口腔天然免疫系統(tǒng)的重要組成部分,與多種口腔感染性疾病密切相關(guān)。研究表明,β-防御素受LPS、TNF-α、IL-1α、IL-1β等炎癥因子的誘導(dǎo)表達(dá)[3-5],而用于治療感染性疾病的中藥能夠增強(qiáng)機(jī)體免疫能力,但其作用機(jī)制尚不明確,把β-防御素作為篩選抗菌中藥或其有效成分作為新的研究方向,可以增加中藥藥效的物質(zhì)基礎(chǔ),甚至還可能為解釋中藥抗感染機(jī)制提供新的依據(jù)[6]。本實(shí)驗(yàn)期望篩選出相關(guān)中藥,可以為β-防御素-2基因表達(dá)的研究提供實(shí)驗(yàn)依據(jù),現(xiàn)報(bào)道如下。
