摘 要:對建立豬和牛的胚胎干細(xì)胞系(Embryonic stem cells,ESCs)的條件進(jìn)行了摸索和優(yōu)化,建立了良好的胚胎培養(yǎng)體系、內(nèi)細(xì)胞團(tuán)分離方法和飼養(yǎng)層的選擇;另外分別建立豬和牛的誘導(dǎo)性多能性干細(xì)胞系(induced pluripotent stem cells,iPS),并分別對其進(jìn)行了鑒定;對目前的培養(yǎng)體系進(jìn)行了優(yōu)化提高了iPS的效率和發(fā)育潛能;嘗試以iPS為供體細(xì)胞進(jìn)行核移植,以期獲得iPS來源的動物種群。

Abstract:Exploration and optimization the conditions for establishment of pigs and cattle embryonic stem cell lines (ESCs) to establish a good embryo culture system, the inner cell mass separation methods and choice of feeder layers. Established pigs and cattle induced pluripotent stem cells (iPSCs), with respect to their identification; the current training system has been optimized to improve the efficiency and growth potential of iPSCs; try to obtain iPS-derived animal populations, by nuclear transfer that iPS cells as donor cells.
