      褪黑素與乳腺癌大鼠外周血Treg相關(guān)細(xì)胞因子的關(guān)系研究

      【摘要】 目的:研究褪黑素與乳腺癌大鼠外周血血清中轉(zhuǎn)化生長因子β1(Transforming growth factor-β1,TGF-β1)和白介素35(Interleukin-35,IL-35)水平的關(guān)系,分析褪黑素對乳腺癌大鼠外周血中調(diào)節(jié)性T細(xì)胞(Regulatory T cell,Treg cell)相關(guān)細(xì)胞因子及腫瘤免疫的影響。方法:選用兩月齡雌性SD大鼠60只,接種大鼠乳腺癌SHZ-88細(xì)胞,按照隨機(jī)數(shù)字表法分成實(shí)驗組(注射褪黑素)和對照組(注射生理鹽水),10 d后乳腺癌大鼠荷瘤動物模型建成,抽取大鼠股動脈外周血,采用ELISA法檢測外周血血清中TGF-β1、IL-35的濃度。結(jié)果:實(shí)驗組乳腺癌大鼠外周血TGF-β1濃度低于對照組,差異有統(tǒng)計學(xué)意義(P<0.05);實(shí)驗組乳腺癌大鼠外周血IL-35濃度低于對照組,差異有統(tǒng)計學(xué)意義(P<0.05)。結(jié)論:褪黑素能降低乳腺癌大鼠外周血中Treg相關(guān)細(xì)胞因子的表達(dá),褪黑素可能具有抑制乳腺癌細(xì)胞免疫逃避從而拮抗乳腺癌進(jìn)展的作用。

      【Abstract】 Objective:To study the relationship between melatonin and expression levels of transforming growth factor-β1 and interleukin-35 in peripheral blood serum of breast cancer rats,and to analyze the effect of melatonin on regulatory T cell related cytokines in peripheral blood and tumor immunity of breast cancer rats.Method:A total of 60 female SD rats in age 2 months were selected,and the rat breast cancer SHZ-88 cells were inoculated in the rats.The rats were randomly divided to the experimental group injected with Melatonin and the control group injected with saline.The tumor bearing animal models of breast cancer rats were established on the 10th day after inoculating.The peripheral blood of rats femoral artery,and the concentrations of TGF-β1 and

      IL-35 in peripheral blood serum were detected by ELISA method.Result:In the experimental group,the concentration of TGF-β1 in peripheral blood of breast cancer rats was lower than that of the control group(P<0.05).In the experimental group,the concentration of IL-35 in peripheral blood of breast cancer rats was lower than that of the control group(P<0.05).Conclusion:Melatonin can reduce the expression of Treg related cytokines in the peripheral blood of breast cancer rats,which may have the effect of inhibiting the immune evasion of breast cancer cells to antagonize the progression of breast cancer.

      乳腺癌的發(fā)生發(fā)展與機(jī)體內(nèi)免疫功能失常,對腫瘤細(xì)胞缺乏識別和殺滅能力,導(dǎo)致腫瘤實(shí)現(xiàn)免疫逃避從而不斷增殖浸潤和遠(yuǎn)處轉(zhuǎn)移密切相關(guān)[1]。已有研究表明機(jī)體內(nèi)Treg細(xì)胞表達(dá)的細(xì)胞因子在維持腫瘤免疫微環(huán)境中起重要作用,其中轉(zhuǎn)化生長因子β1(TGF-β1)和白介素35(IL-35)對惡性腫瘤的進(jìn)展具有促進(jìn)作用[2-3]。褪黑素是由松果體分泌的具有調(diào)控機(jī)體生物節(jié)律、抗氧化、抗腫瘤作用的神經(jīng)內(nèi)分泌激素,是否能影響惡性腫瘤中Treg相關(guān)細(xì)胞因子表達(dá)尚待研究[4-5]。本研究通過建立乳腺癌大鼠荷瘤動物模型,研究褪黑素與乳腺癌大鼠外周血中TGF-β1和IL-35的關(guān)系,探討褪黑素對乳腺癌中Treg相關(guān)細(xì)胞因子和腫瘤免疫的影響。

      1 材料與方法

      1.1 實(shí)驗動物和試劑 選用兩月齡雌性SD大鼠60只,體重范圍(200±10)g,標(biāo)準(zhǔn)飼養(yǎng)。按照隨機(jī)數(shù)字表法分為兩組,分別為注射生理鹽水的對照組和按15 mg/(kg·d)注射褪黑素的實(shí)驗組,每組30只。大鼠乳腺癌SHZ-88細(xì)胞購于上海生命科學(xué)院,褪黑素購于美國Sigma公司,TGF-β1和IL-35的ELISA試劑盒購于美國R&D公司。

      1.2 建立動物模型 乳腺癌細(xì)胞株于培養(yǎng)瓶中傳代培養(yǎng),當(dāng)癌細(xì)胞培養(yǎng)至對數(shù)生長期時,吹打制成單細(xì)胞懸液,每只大鼠按5×105個細(xì)胞量在大腿內(nèi)側(cè)根部皮下接種SHZ-88乳腺癌單細(xì)胞懸液,

      1次/只。自接種日起,每日18∶00分別按對照組和實(shí)驗組的不同干預(yù)手段,給予腹腔內(nèi)注射生理鹽水或15 mg/(kg·d)的褪黑素,1次/d。接種后第10天可在接種處觸及皮下瘤塊,抽取另一側(cè)大腿股動脈外周血待檢測,處死荷乳腺癌大鼠,切除接種處瘤塊并經(jīng)病理切片觀察證實(shí)為乳腺癌。

      1.3 ELISA法檢測 采用雙抗體夾心ELISA法檢測大鼠外周血血清中TGF-β1、IL-35的水平,操作嚴(yán)格按照ELISA試劑盒說明書,檢測過程設(shè)置標(biāo)準(zhǔn)品和空白對照,根據(jù)標(biāo)準(zhǔn)曲線計算出樣品中TGF-β1、IL-35的濃度。

      1.4 統(tǒng)計學(xué)處理 采用SPSS 20.0軟件對所得數(shù)據(jù)進(jìn)行統(tǒng)計分析,計量資料用(x±s)表示,比較采用t檢驗,P<0.05為差異有統(tǒng)計學(xué)意義。

      2 結(jié)果

      2.1 兩組外周血中TGF-β1水平比較 實(shí)驗組(注射褪黑素)乳腺癌大鼠外周血TGF-β1濃度為(132.4±25.7)pg/mL,低于對照組(注射生理鹽水)乳腺癌大鼠外周血TGF-β1的(157.7±40.6)pg/mL,差異有統(tǒng)計學(xué)意義(t=2.8839,P<0.01)。

      2.2 兩組外周血中IL-35水平比較 實(shí)驗組(注射褪黑素)乳腺癌大鼠外周血IL-35濃度為(95.8±23.2)pg/mL,低于對照組(注射生理鹽水)乳腺癌大鼠外周血IL-35的(112.2±29.7)pg/mL,差異有統(tǒng)計學(xué)意義(t=2.3835,P<0.05)。

      3 討論

      褪黑素是由松果體分泌的一種生物活性物質(zhì),屬于吲哚類神經(jīng)內(nèi)分泌激素[6]。褪黑素具有高度脂溶性和水溶性,易透過細(xì)胞膜,主要在肝臟內(nèi)代謝轉(zhuǎn)化為多功能的吲哚胺而發(fā)揮生物學(xué)效應(yīng),另有少量經(jīng)腎臟排泄,褪黑素半衰期短,不易蓄積,因此毒性很小[7]。褪黑素具有多種生理功能,呈晝夜波動性分泌,具有調(diào)節(jié)機(jī)體生物節(jié)律、激素分泌、免疫功能的作用,尤其是神經(jīng)-內(nèi)分泌-免疫調(diào)節(jié)方面功能的異常與惡性腫瘤的發(fā)生發(fā)展密切相關(guān)[8]。Vigoré等[9]研究表明褪黑素能通過抑制Treg細(xì)胞相關(guān)免疫效應(yīng)拮抗惡性腫瘤進(jìn)展,但其機(jī)制尚未明確。Treg細(xì)胞是具有免疫抑制功能的T細(xì)胞亞群,在維持機(jī)體免疫耐受,抑制機(jī)體過度免疫反應(yīng),誘導(dǎo)腫瘤免疫逃避等方面具有極其重要的作用;相關(guān)研究表明,多種惡性腫瘤中Treg細(xì)胞產(chǎn)生的數(shù)量增多,且與腫瘤的惡性度、分期呈正相關(guān),而與生存預(yù)后負(fù)相關(guān)[10-11]。關(guān)于Treg細(xì)胞發(fā)揮其免疫抑制作用的機(jī)制,除了與樹突狀細(xì)胞(DC)直接結(jié)合抑制DC和效應(yīng)T細(xì)胞功能外,還有通過分泌Treg相關(guān)的抑制性細(xì)胞因子抑制機(jī)體免疫的途徑。在惡性腫瘤發(fā)生發(fā)展過程中TGF-β1和IL-35是非常重要的Treg相關(guān)細(xì)胞因子[12-13]。

      轉(zhuǎn)化生長因子β1基因定位于染色體19q13.1,由兩條相同多肽鏈經(jīng)二硫鍵相連構(gòu)成二聚體,TGF-β1是轉(zhuǎn)化生長因子超家族中具有代表性的分子,主要由Treg細(xì)胞表達(dá),與腫瘤的進(jìn)展關(guān)系密切[14-15]。TGF-β1發(fā)揮促腫瘤生長的機(jī)制除了通過激活絲裂原活化蛋白激酶通路促進(jìn)腫瘤細(xì)胞增殖,還能負(fù)調(diào)控免疫活性細(xì)胞如自然殺傷細(xì)胞和單核細(xì)胞等導(dǎo)致機(jī)體對腫瘤的免疫作用受到抑制,應(yīng)用TGF-β1結(jié)合蛋白、TGF-β1抗體或反義

      TGF-β1 mRNA可以中和TGF-β1的作用,拮抗腫瘤的侵襲轉(zhuǎn)移[14-16]。本研究結(jié)果顯示,注射褪黑素的荷乳腺癌大鼠外周血中TGF-β1濃度顯著低于注射生理鹽水的對照組荷瘤大鼠,表明褪黑素能有效降低Treg細(xì)胞的TGF-β1表達(dá)量,抑制TGF-β1的促腫瘤生長作用,提示褪黑素能通過下調(diào)Treg相關(guān)細(xì)胞因子表達(dá)的途徑,發(fā)揮其對乳腺癌發(fā)生發(fā)展的抑制作用。

      白介素35是1997年Devergne等[17]發(fā)現(xiàn)的一種新型抗炎細(xì)胞因子EBI3-p35,屬于IL-12家族,由Treg細(xì)胞特異性產(chǎn)生,不僅能抑制效應(yīng)T細(xì)胞,還能誘導(dǎo)T細(xì)胞轉(zhuǎn)化為更具有抑制活性的調(diào)節(jié)性

      T細(xì)胞(iTr35)。正常情況下,機(jī)體Treg細(xì)胞不會產(chǎn)生IL-35,但在發(fā)生自身免疫性疾病、惡性腫瘤等情況時,其表達(dá)量上升[18-19]。IL-35可通過增強(qiáng)腫瘤細(xì)胞免疫逃避能力、提高腫瘤細(xì)胞增殖能力等作用促進(jìn)腫瘤的侵襲轉(zhuǎn)移[20-21]。本研究結(jié)果顯示,注射褪黑素的荷乳腺癌大鼠外周血中IL-35濃度顯著低于注射生理鹽水的對照組荷瘤大鼠,提示褪黑素能使Treg細(xì)胞顯著減少IL-35的表達(dá),改變Treg細(xì)胞相關(guān)的乳腺癌免疫微環(huán)境,降低Treg相關(guān)細(xì)胞因子介導(dǎo)的腫瘤免疫逃避能力,發(fā)揮褪黑素的抗乳腺癌作用。

      褪黑素具有抑制乳腺癌機(jī)體外周血中Treg相關(guān)細(xì)胞因子表達(dá)的作用,可以拮抗Treg細(xì)胞的免疫抑制功能,減少乳腺癌細(xì)胞的免疫逃避,增強(qiáng)機(jī)體的抗腫瘤免疫反應(yīng),從而控制腫瘤的進(jìn)展,因此褪黑素治療有望成為抗乳腺癌治療的新途徑之一。
