粉葛為豆科葛屬植物甘葛藤PuerariathomsoniiBenth.的干燥根[1],在全國(guó)分布廣泛,主產(chǎn)地為廣西、廣東、江西、云南、四川等地,并且在老撾、泰國(guó)、緬甸、不丹等亞洲國(guó)家也均有分布[2],收載于2020年版《中國(guó)藥典》,是常用藥食同源中藥,具有解肌退熱、生津止渴、通經(jīng)活絡(luò)、解酒毒等功效,用于治療外感發(fā)熱頭痛、項(xiàng)背強(qiáng)痛、眩暈頭痛、中風(fēng)偏癱、胸痹心痛等疾病[1],與葛根功能主治完全相同,并且食用歷史悠久,淀粉含量高,市場(chǎng)需求大,是制作葛粉的主要原料,也可作雜食食用。
目前,國(guó)內(nèi)外學(xué)者對(duì)同屬植物葛根的藥效物質(zhì)及作用機(jī)制的關(guān)注較多。越來(lái)越多的研究表明,葛根、粉葛功能主治雖然相同,但兩者藥效物質(zhì)及臨床使用具有一定差異[3-5],對(duì)粉葛藥效物質(zhì)基礎(chǔ)的研究較少,僅Arao等[6]從中分離得到5個(gè)三萜皂苷類成分,而且大多集中于葛根素含量測(cè)定或質(zhì)量控制。因此,采用現(xiàn)代色譜分離技術(shù)方法對(duì)粉葛進(jìn)行系統(tǒng)化學(xué)成分研究,從中分離鑒定出29個(gè)化合物,其中11個(gè)為首次從該屬植物中分離得到。
Avance Ⅲ 600型核磁共振儀(以TMS為內(nèi)標(biāo),德國(guó)Bruker公司);TripleTOF5600液相色譜高分辨串聯(lián)質(zhì)譜儀(ESI-MS,美國(guó)AB公司);LC-3000型制備高效液相色譜儀(北京創(chuàng)新通恒科技有限公司);安捷倫1260分析高效液相色譜儀、安捷倫1200制備高效液相色譜儀(美國(guó)安捷倫公司)。制備型色譜柱(250 mm×21.2 mm,10 μm) (美國(guó)Phenomenex公司);Sepax GP-Phenyl (250 mm×10 mm,10 μm) 半制備型色譜柱(蘇州賽分科技股份有限公司);硅膠G(100~200、200~300目,青島海洋化工有限公司);MCI (CHP-20P,日本三菱化學(xué)公司);凝膠(Sephadex LH-20,瑞士Pharmacia Biotech公司);反相硅膠C18(50 μm,日本富士公司)。粉葛購(gòu)于江西江中中藥飲片有限公司(產(chǎn)地廣西玉林),經(jīng)江西中醫(yī)藥大學(xué)藥學(xué)院葛菲教授鑒定為豆科葛屬植物甘葛藤PuerariathomsoniiBenth.的干燥根。半制備及制備液相所用試劑為色譜純;其他均為分析純。
取飲片45.0 kg,粉碎,乙醇加熱回流提取3次,濾過,合并濾液,減壓回收溶劑,得浸膏3.2 kg,加入10 L純凈水混懸,依次用等體積石油醚、乙酸乙酯、正丁醇各萃取3次,回收有機(jī)溶劑,分別得石油醚部位、乙酸乙酯部位0.18 kg、正丁醇部位1.28 kg、水部位。
乙酸乙酯部位分別用石油醚-丙酮(65∶35)、乙酸乙酯-甲醇(50∶50)進(jìn)行硅膠柱層析,薄層色譜檢測(cè)合并相同流分,得Fr.1 (59.6 g)、Fr.2 (74.0 g)、Fr.3 (71.3 g)、Fr.4 (27.9 g)。Fr.1經(jīng)Sephadex LH-20凝膠柱,二氯甲烷∶甲醇(1∶1)洗脫,得流分Fr.1.1~Fr.1.5,F(xiàn)r.1.3經(jīng)制備反相HPLC,乙腈-水 (25∶75~55∶45)洗脫,得流分Fr.1.31~Fr.1.38,F(xiàn)r.1.34經(jīng)半制備反向HPLC,乙腈-水(20∶80)洗脫,得到化合物1(6.5 mg);Fr.1.35經(jīng)半制備反向HPLC,乙腈-水(35∶65)洗脫,得到化合物2(7.0 mg)、3(6.3 mg)、4(7.1 mg);Fr.1.37經(jīng)半制備反向HPLC,乙腈-水(45∶55)洗脫,得到化合物5(7.7 mg)。Fr.2~Fr.4依次經(jīng)Sephadex LH-20凝膠柱色譜脫色、制備反相HPLC分離、半制備反相HPLC純化,乙腈-水(40∶60~70∶30)洗脫。最終,從Fr.2中分離得到9個(gè)化合物,分別為6(8.0 mg)、7(6.1 mg)、8(7.5 mg)、9(7.0 mg)、10(6.8 mg)、11(6.5 mg)、12(8.5 mg)、14(7.2 mg)、13(7.0 mg);從Fr.3中分離得到2個(gè)化合物,分別為15(8.0 mg)、16(8.9 mg);從Fr.4中分離得到化合物17(8.2 mg)。
正丁醇部位經(jīng)硅膠柱,乙酸乙酯-甲醇(40∶60~20∶80)洗脫,TLC分析合并相同流分,得到Fr.5(99.3 g)、Fr.6(700.0 g)、Fr.7(163.7 g)、Fr.8(39.0 g)。Fr.5~Fr.7依次經(jīng)Sephadex LH-20凝膠柱脫色、制備反相HPLC分離、半制備反相HPLC純化,甲醇-水 (30∶70~70∶30)洗脫。最終,從Fr.5中分離得到8個(gè)化合物,分別為18(6.0 mg)、19(7.5 mg)、20(7.8 mg)、21(6.4 mg);從Fr.6中分離得到4個(gè)化合物,分別為22(8.3 mg)、23(7.0 mg)、24(9.0 mg)、25(9.5 mg);從Fr.7中分離得到4個(gè)化合物,分別為26(6.7 mg)、27(8.0 mg)、28(8.1 mg)、29(10.8 mg)。
化合物1:無(wú)色晶體,分子式C10H8O3,ESI-MSm/z: 177.054 5[M+H]+。1H-NMR (CD3OD, 600 MHz)δ:7.80 (2H, dd,J=7.8, 1.9 Hz, H-2′,6′), 7.53~7.45 (3H, m, H-3′,4′,5′), 6.58 (1H, s,H-4), 6.54 (1H, s,H-2);13C-NMR (CD3OD, 150 MHz)δ:173.4 (C-1), 115.6 (C-2), 165.4 (C-3), 99.9 (C-4), 131.0 (C-1′), 129.2 (C-2′,6′), 130.0 (C-3′,5′),132.5 (C-4′)。以上數(shù)據(jù)與文獻(xiàn)[7]報(bào)道基本一致,鑒定為5-hydroxy-4-phenyl-2(5H)-furanone。

采用正相和反相硅膠柱色譜、凝膠柱色譜、反相高效液相色譜等分離純化方法,結(jié)合核磁、質(zhì)譜等波譜技術(shù),對(duì)粉葛進(jìn)行化學(xué)成分研究,結(jié)果從中分離鑒定出29個(gè)化合物,包括8個(gè)異黃酮苷元(5、6、8、11、2、14、16、29)、10個(gè)異黃酮苷(15、17、19、20、22~25、27、28)、5個(gè)有機(jī)酸酯(2~4、13、21)、2個(gè)氨基酸、4個(gè)黃烷酮等其他成分,其中化合物1、2、4、7、9、11、13、18、19、21、26為首次從葛屬植物中分離得到。
葛屬植物所含成分的結(jié)構(gòu)類型主要為異黃酮,葛根、粉葛主要成分類型相同,均為葛根素、大豆苷等異黃酮類,但前者含有葛酚苷類特征性成分,而后者目前暫未發(fā)現(xiàn)[5]。另外,葛根、粉葛的藥典標(biāo)準(zhǔn)除葛根素含量的要求不同外,功效主治、主要化學(xué)成分結(jié)構(gòu)類型和含量均相同,但兩者成分種類也存在一些差異,是否可以此為依據(jù)進(jìn)行區(qū)別尚不明確,有待進(jìn)一步研究。
