脈花黨參(Codonopsis nervosa)為桔梗科(Ganpanulaceae)黨參屬草本,產(chǎn)于我國(guó)西藏東部、青海東南部、四川西部、云南西北部及甘肅東南部等地,是藏藥陸得多吉的原植物之一,性苦、幸、澀、涼,具有消炎散腫、滋陰壯陽(yáng)的功效,在我國(guó)藏族中常用于關(guān)節(jié)炎、神經(jīng)痛、麻風(fēng)、腳氣病及癔病的治療[1]。到目前為止,未見(jiàn)對(duì)該植物化學(xué)成分的報(bào)道,為了尋找新的活性成分及為藏藥陸得多吉質(zhì)量標(biāo)準(zhǔn)的制定提供依據(jù),我們對(duì)脈花黨參帶根全草的化學(xué)成分進(jìn)行了研究,從乙酸乙酯部分分離鑒定了6種成分:羽扇豆醇(1),β-谷甾醇(2),何伯烷-6α,22-二醇(3),山柰酚(4),槲皮素(5),獐牙菜苷(6)。以上化合物均為首次從該植物中分離得到。
藥材采自甘肅文縣,經(jīng)西北民族大學(xué)化工學(xué)院范文革副教授鑒定為脈花黨參(Codonopsis nervosa)的全草。
核磁共振譜用Broker AM-400型核磁共振儀測(cè)定;EIMS質(zhì)譜用MAT-7ll型質(zhì)譜儀測(cè)定;薄層色譜及柱色譜所用硅膠(200~300目)均為中國(guó)青島海洋化工集團(tuán)公司生產(chǎn);Sephadex LH-20、ODS為Pharmacia Sweden公司生產(chǎn);實(shí)驗(yàn)試劑均為分析純;熔點(diǎn)用Buchi 510熔點(diǎn)儀測(cè)定(溫度未校正)。
脈花黨參全草(3.0 kg)經(jīng)過(guò)粉粹后,95%EtOH回流提取3次,提取液合并減壓濃縮得到浸膏260 g。浸膏用熱水懸浮,依次用石油醚,乙酸乙酯,正丁醇萃取。乙酸乙酯部分(42 g)用石油醚-乙酸乙酯體系(20 ∶1,10 ∶1,8 ∶1,6 ∶1,4∶1,3∶1,2∶1,1 ∶1梯度洗脫)進(jìn)行硅膠柱層析,分段收集,得到8個(gè)部分(CN-1-CN-8)。CN-3經(jīng)硅膠反復(fù)柱層析純化,得化合物1(50 mg)、2(150 mg)、CN-4經(jīng)Sephadex LH-20及硅膠反復(fù)柱層析純化,得化合物3(9 mg)、CN-6經(jīng)Sephadex LH-20及硅膠反復(fù)柱層析純化,得化合物4(20 mg),CN-8經(jīng)Sephadex LH-20、ODS及硅膠反復(fù)柱層析純化,得化合物5(10 mg)、6(12 mg)。
化合物1:白色針狀結(jié)晶,mp 210~212℃。1H-NMR(CDCl3,400 MHz,δ):3.21(1H,dd,J=10.6,3.0 Hz,H-3),2.38(1H,m,H-19),4.65(1H,br s,H-29a),4.56(1H,br s,H-29b),1.73(3H,s,H-30),1.02(3H,s,H-26),0.95(3H,s,H-27),0.97(3H,s,H-23),0.83(3H,s,H-25),0.79(3H,s,H-28),0.76(3H,s,H-24)。其波譜數(shù)據(jù)與文獻(xiàn)[2]報(bào)道的羽扇豆醇一致,與羽扇豆醇對(duì)照品對(duì)照多種溶劑系統(tǒng)展開(kāi)Rf值一致,混合熔點(diǎn)不下降,故鑒定為羽扇豆醇。
化合物2:無(wú)色針狀結(jié)晶,mp136~137℃。與β-谷甾醇對(duì)照品對(duì)照多種溶劑展板Rf值一致,混合熔點(diǎn)不下降。故鑒定為β-谷甾醇。
化合物3:無(wú)色粒狀結(jié)晶(丙酮),mp 223~225℃;1HNMR(C5D5N,400 MHz,δ):3.95(1H,td,J=9.8,4.6 Hz,H-6),0.76,0.85,0.97,1.01,1.03,1.15,1.18,1.21(均 3H,s,H-23-H-30);13C-NMR(C5D5N,100 MHz,δ):40.66(C-1),18.97(C-2),44.29(C-3),34.13(C-4),61.18(C-5),67.89(C-6),45.78(C-7),44.20(C-8),49.72(C-9),40.66(C-10),21.35(C-11),24.36(C-12),50.18(C-13),42.88(C-14),34.65(C-15),22.27(C-16),54.60(C-17),44.29(C-18),41.66(C-19),26.98(C-20),51.50(C-21),72.42(C-22),37.30(C-23),22.52(C-24),17.28(C-25),18.49(C-26),17.40(C-27),16.41(C-28),29.78(C-29),31.45(C-30)。其波譜數(shù)據(jù)與文獻(xiàn)報(bào)道的何伯烷-6α,22-二醇的數(shù)據(jù)一致[3],因此鑒定為何伯烷-6α,22-二醇。
化合物4:黃色針狀晶體(甲醇),mp 275~277℃。1HNMR(CD3COCD3,400 MHz,δ):7.01(2H,d,J=8.6 Hz,H-3,,5,),8.15(2H,d,J=8.6 Hz,H-2′,6′),6.28(1H,d,J=2.0 Hz,H-6),6.55(1 H,d,J=2.0 Hz,H-8)。13C-NMR(CD3COCD3,100 MHz,δ):146.13(C-2),135.16(C-3),175.17(C-4),156.11(C-5),98.24(C-6),163.89(C-7),93.34(C-8),160.21(C-9),103.12(C-10),121.88(C-1′),129.24(C-2′),115.16(C-3′),159.14(C-4′),115.15(C-5′),129.24(C-6′)。以上數(shù)據(jù)與文獻(xiàn)[4]報(bào)道一致,確定該化合物為山柰酚(Kaempfero1)。
化合物5:黃色粉末狀結(jié)晶,mp 313~314℃。1H-NMR(CD3SOCD3,400 MHz,δ):12.49(1H,s,5-OH),10.86(1H,s,7-OH),9.64(1H,s,3-OH),9.40(1H,s,4′-OH),9.36(1H,s,3′-OH),7.54(1H,d,J=8.0 Hz,H-6′),7.78(1H,s,H-2′),6.89(1H,d,J=8.0 Hz,H-5′),6.43(1H,s,H-8),6.19(1H,s,H-6),其熔點(diǎn)及波譜數(shù)據(jù)與文獻(xiàn)[5]報(bào)道的槲皮素一致,與槲皮素對(duì)照品對(duì)照多種溶劑系統(tǒng)展開(kāi)Rf值一致,故鑒定為槲皮素。
化合物 6:白色粉末狀固體。1H-NMR(CD3OD,400 MHz,δ):5.56(1H,d,J=1.6 Hz,H-1),7.61(1H,d,J=2.3 Hz,H-3),3.11-3.46(5H,m,H-2′-H-5′,H-5),1.75(1H,m,H-6a),1.62(1H,m,H-6b),4.34-4.44(2H,m,H-7),5.56(1H,m,H-8),2.70(1H,m,H-9),5.26(1H,m,H-10a),5.31(2H,m,H-10b),4.67(1H,d,J=7.8 Hz,H-1′),3.90(1H,dd,J=12.2,1.2 Hz,H-6′),3.65(1H,dd,J=12.2,6.8 Hz,H-6′)。13C-NMR(CD3COD,100 MHz,δ):98.40(C-1),154.42(C-3),106.46(C-4),28.82(C-5),26.31(C-6),70.17(C-7),133.76(C-8),44.16(C-9),121.35(C-10),100.10(C-1′),75.09(C-2′),78.24(C-3′),71.90(C-4′),78.74(C-5′),63.08(C-6′),168.95(C=O),其波譜數(shù)據(jù)與獐牙菜苷(sweroside)的數(shù)據(jù)一致[6],故鑒定為獐牙菜苷。
