動(dòng)物生產(chǎn)層鹽藻粉在奶牛生產(chǎn)中的應(yīng)用

摘要:鹽藻(Dunaliella salina)是一種單細(xì)胞無(wú)細(xì)胞壁的真核生物,主要用于提煉天然β-胡蘿卜素和多糖。鹽藻粉是鹽藻干燥后獲得的粉狀顆粒,具有抗氧化、抗病毒、抗腫瘤和提高免疫力等生物學(xué)活性。目前,鹽藻粉在人類和嚙齒類動(dòng)物上的研究較多,而其在奶牛生產(chǎn)中的應(yīng)用很少。本文從瘤胃發(fā)酵、氧化應(yīng)激調(diào)控、免疫功能、營(yíng)養(yǎng)素利用和生產(chǎn)性能5個(gè)方面綜述了鹽藻粉對(duì)奶牛代謝的影響和潛在應(yīng)用價(jià)值,以期為鹽藻粉在奶牛生產(chǎn)中的應(yīng)用提供參考依據(jù)。

鹽藻(Dunaliellasalina)是一類極耐鹽的單細(xì)胞無(wú)細(xì)胞壁單倍體綠藻。19世紀(jì)30年代,法國(guó)人達(dá)納爾首次在地中海沿岸的鹽池中發(fā)現(xiàn),因其缺乏細(xì)胞壁,與其它雙鞭毛單細(xì)胞藻不同,故后人將其命名為新屬Dunaliella[1]。鹽藻屬于綠藻門(Chlorphyta),綠藻綱(Ehlorphyeeae),團(tuán)藻目(Volvoeales),杜氏藻科(Dunalielljaceae)。鹽藻是光合自養(yǎng)型真核生物,具有繁殖快、適應(yīng)性廣和抗逆性強(qiáng)等特點(diǎn)[2-3]。因此,其當(dāng)前作為耐鹽機(jī)理研究的模式生物[4]、生物遺傳的效應(yīng)器[5]和新型綠色的生物能源[6],還被用于天然β-胡蘿卜素、多糖、藥材、食品和動(dòng)物飼料等的工廠化生產(chǎn)[7]。
1鹽藻的概述
1.1鹽藻的營(yíng)養(yǎng)價(jià)值
鹽藻含有豐富的蛋白質(zhì)、不飽和脂肪酸、多糖、維生素和18種氨基酸等元素[8]。此外,鹽藻中無(wú)機(jī)元素含量高,比例適宜,可作為人體無(wú)機(jī)元素的補(bǔ)充劑[9]。正常生長(zhǎng)的鹽藻細(xì)胞中,蛋白質(zhì)含量占細(xì)胞干重50%~60%[9],碳水化合物占12%~40%[10-11],脂類占6%~10%[12]。鹽藻是世界上含天然β-胡蘿卜素較高的生物之一[13-15]。尤其是順式β-胡蘿卜素,具有防輻射、延緩機(jī)體衰老、增強(qiáng)淋巴細(xì)胞活性、增強(qiáng)機(jī)體免疫力、抑制腫瘤和防癌抗癌等作用。由其制備的鹽藻粉無(wú)毒[16],且仍能保持良好的營(yíng)養(yǎng)成分。研究表明,鹽藻粉中蛋白質(zhì)、多糖和天然β-胡蘿卜素含量分別為15%~30%、10%和12%,其中順式β-胡蘿卜素占天然β-胡蘿卜素總量的50%[17]。其中氨基酸種類齊全,限制性氨基酸含量高(賴氨酸含量791.8 mg·100 g-1)[17]。
1.2鹽藻粉的生產(chǎn)與應(yīng)用
鹽藻粉的生產(chǎn)工藝包括對(duì)鹽藻的選種、養(yǎng)殖、采收、分離和干燥等過(guò)程[18]。鹽藻粉制備要點(diǎn)在于鹽藻的采收[19]。由于鹽藻的比重與培養(yǎng)液相近,因此,加大了分離純化的難度,根據(jù)生產(chǎn)規(guī)模主要分為離心和氣浮兩種分離方式[20]。鹽藻粉的生產(chǎn)力求做到簡(jiǎn)單、低耗、高效且不破壞營(yíng)養(yǎng)成分[1]。研究表明,鹽藻粉可促進(jìn)動(dòng)物生長(zhǎng),提高飼料轉(zhuǎn)化率和生產(chǎn)性能,減少疾病的發(fā)生[21]。
2鹽藻粉對(duì)奶牛營(yíng)養(yǎng)生理的調(diào)控
2.1瘤胃發(fā)酵
高義彪等[22]通過(guò)體外研究發(fā)現(xiàn),添加9 mg鹽藻粉的試驗(yàn)組體外發(fā)酵的各項(xiàng)指標(biāo)表現(xiàn)良好,發(fā)酵液pH(6.48)維持在最適pH范圍,且與對(duì)照組相比揮發(fā)性脂肪酸、氨態(tài)氮和微生物蛋白的含量顯著增加(P<0.05),總產(chǎn)氣量顯著降低(P<0.05)。劉大程等[23]的研究結(jié)果與其相似,鹽藻粉蛋白質(zhì)含量高,品質(zhì)優(yōu),是微生物蛋白的良好來(lái)源。因此,添加鹽藻粉能夠維持瘤胃內(nèi)環(huán)境的穩(wěn)態(tài),并提高營(yíng)養(yǎng)物質(zhì)利用率,促進(jìn)瘤胃發(fā)酵。
2.2氧化應(yīng)激和免疫功能
鹽藻粉中的β-胡蘿卜素具有很強(qiáng)的抗氧化能力,可清除自由基,維持機(jī)體自由基動(dòng)態(tài)平衡[24-25]。因此,奶牛日糧中添加鹽藻粉可能具有緩解氧化應(yīng)激[26-27]和增進(jìn)健康的作用[28]。
趙鵬等[29]試驗(yàn)結(jié)果表明,奶牛(泌乳中期)日糧中添加9 g·d-1的鹽藻粉(β-胡蘿卜素含量為2.4%)可顯著增加血清免疫球蛋白G(Immunoglobulin G, IgG)和免疫球蛋白M(Immunoglobulin M, IgM)抗體含量(P<0.05)。這對(duì)處于特殊生理階段的奶牛,尤其在圍產(chǎn)期[30-31],具有提高母體免疫力[32-33]的重要作用。該免疫調(diào)節(jié)機(jī)理可能與多糖有關(guān)[34],多糖可促進(jìn)抗體的生成與活化。鹽藻中的多糖主要以硫酸基糖蛋白和己糖醛酸糖蛋白的形式存在,參與細(xì)胞的信號(hào)轉(zhuǎn)導(dǎo)[35]。在小鼠[36]和人白細(xì)胞[37]上的研究發(fā)現(xiàn),鹽藻粉對(duì)敗血癥病毒有明顯的抑制效果,主要是硫酸化糖發(fā)揮的抗腫瘤和抗病毒作用[38]。因此,日糧中添加鹽藻粉在一定程度上可增強(qiáng)奶牛的免疫功能。
2.3營(yíng)養(yǎng)素利用
2.3.1鈣和磷Ca和P屬于奶牛的必需常量礦物元素[39],Ca穩(wěn)態(tài)[40]有助于避免產(chǎn)后癱瘓的發(fā)生[41],且Ca是體內(nèi)多種酶的激活劑,參與多種代謝活動(dòng)。P是細(xì)胞膜的重要組成元素,磷脂雙分子層對(duì)多種營(yíng)養(yǎng)素的吸收具有重要作用[42]。因此,適宜的Ca、P含量和比例可維持奶牛正常的生理代謝,提高其生產(chǎn)性能[43]。
宋范成和閆素梅[44]研究發(fā)現(xiàn)(表1),日糧添加鹽藻粉對(duì)Ca和P的表觀消化率無(wú)顯著影響(P>0.05),但隨時(shí)間的延長(zhǎng),Ca和P的表觀消化率均有上升的趨勢(shì),而其作用機(jī)理還不明確。

2.3.2氮隨著奶業(yè)發(fā)展,N排放問(wèn)題備受人們關(guān)注[45]。動(dòng)物產(chǎn)生的N一部分以氣體形式釋放,其中胺、一氧化氮、一氧化二氮和氨氣等有毒氣體可造成空氣污染[46]。另一部分以糞尿的形式排出,可造成水體富營(yíng)養(yǎng)化和酸雨等災(zāi)害[47],威脅人類的健康。當(dāng)其滲入土壤中,會(huì)使土地板結(jié),肥力下降,甚至造成土地荒漠化[48];N排放同時(shí)也造成了能量的損失[49],因此減少N排放迫在眉睫。
研究表明,鹽藻粉和維生素A均可提高日糧N的表觀消化率(P<0.05)[50]。而鹽藻粉富含的β-胡蘿卜素,在動(dòng)物體內(nèi)可轉(zhuǎn)化為VA,因此,N表觀消化率的提高可能與其含有較高的β-胡蘿卜素有關(guān),其作用機(jī)制還有待進(jìn)一步的研究。
3鹽藻粉對(duì)奶牛生產(chǎn)性能和乳成分的影響
3.1奶產(chǎn)量
馬吉峰等[51]研究發(fā)現(xiàn),在整個(gè)試驗(yàn)期添加4.5、9.0和13.5 mg鹽藻粉使奶牛奶產(chǎn)量分別提高了3.30%、5.25%和7.05%。趙鵬等[29]研究也發(fā)現(xiàn),添加鹽藻粉組的奶牛日平均奶產(chǎn)量與對(duì)照組相比提高了10.54%,試驗(yàn)牛均處于泌乳中后期,泌乳量呈下降趨勢(shì),但添加鹽藻粉的試驗(yàn)組產(chǎn)奶量下降幅度明顯小于對(duì)照組。因此,奶牛日糧中添加鹽藻粉具有提高奶產(chǎn)量的趨勢(shì)。
3.2乳成分
乳蛋白和乳脂率決定牛奶的價(jià)格[51]。一些主要的乳品生產(chǎn)國(guó)家,如意大利、美國(guó)和法國(guó)生乳脂肪含量標(biāo)準(zhǔn)分別為>3.7%、>3.5%、>3.8%[52],而我國(guó)新的國(guó)標(biāo)GB19301-2010《生乳》中脂肪含量標(biāo)準(zhǔn)為>3.1%。研究表明,意大利、美國(guó)和法國(guó)乳奶白的含量均在3.2%以上[53],但我國(guó)的標(biāo)準(zhǔn)卻從1986年的2.95%降低到2.80%。我國(guó)的乳品質(zhì)與國(guó)外相比還有一定的差距。
馬吉峰等[51]研究表明(表2),隨著鹽藻粉添加量的增加,乳蛋白、乳脂率和乳糖含量均呈上升的趨勢(shì),9 mg試驗(yàn)組與對(duì)照組相比,乳蛋白、乳脂率和乳糖分別提高了2.9%、1.1%和1.2%。因此,添加鹽藻粉可以提高乳蛋白、乳脂率和乳糖含量。關(guān)于鹽藻粉對(duì)奶牛乳腺細(xì)胞泌乳功能的調(diào)控還未見(jiàn)報(bào)道,且其有效成分也尚未明確。

3.3體細(xì)胞數(shù)
測(cè)定鮮牛奶中的體細(xì)胞數(shù),是判斷乳房炎有無(wú)和輕重程度的有力手段[54-55]。牛奶中的體細(xì)胞參加免疫調(diào)節(jié)對(duì)抗病原,當(dāng)微生物侵入時(shí),導(dǎo)致乳房局部血流量增加[56],血管通透性增強(qiáng),造成中性白細(xì)胞和單核細(xì)胞等白血球在有炎癥的血管局部游離,最終脫落進(jìn)入乳汁[57]。一般認(rèn)為體細(xì)胞數(shù)低于2.5×105CFU·mL-1的乳區(qū)是無(wú)炎癥乳區(qū);在2.5×105~1.0×106CFU·mL-1之間,則說(shuō)明奶?;加须[性乳房炎;一旦計(jì)數(shù)超過(guò)1.0×106CFU·mL-1,奶牛就會(huì)有發(fā)生急性炎癥的可能[58]。體細(xì)胞數(shù)的多少嚴(yán)重影響乳品質(zhì),歐盟、新西蘭和澳大利亞等發(fā)達(dá)國(guó)家已明確指出,收購(gòu)生乳的體細(xì)胞數(shù)必須小于4×105CFU·mL-1[59],我國(guó)對(duì)鮮乳中體細(xì)胞數(shù)逐漸重視起來(lái),已有部分乳品公司在收購(gòu)生乳時(shí)開(kāi)始控制體細(xì)胞數(shù)。
馬吉峰等[51]的試驗(yàn)中添加4.5、9.0和13.5 mg鹽藻粉的試驗(yàn)組較對(duì)照組的牛奶體細(xì)胞數(shù)分別降低了15.7%、29.4%和33.3%(表2)。趙鵬等[29]的試驗(yàn)也說(shuō)明,添加鹽藻粉能夠顯著降低體細(xì)胞數(shù)。由此可見(jiàn),日糧添加鹽藻粉可降低體細(xì)胞數(shù),保障乳腺健康和提高牛奶品質(zhì)。
綜上所述,鹽藻粉對(duì)提高奶牛的生產(chǎn)性能有積極作用,原因有以下幾點(diǎn):藻粉蛋白質(zhì)含量高,促進(jìn)微生物蛋白的合成,增加過(guò)瘤胃蛋白含量,提高產(chǎn)奶量,乳脂率和乳糖含量。Higginbotham等[59]和Taylor等[60]研究表明,奶牛日糧中過(guò)瘤胃蛋白水平提高,產(chǎn)奶量、乳脂率和乳糖含量增加,可為此提供一定科學(xué)依據(jù);藻粉中大量的β-胡蘿卜素對(duì)乳腺細(xì)胞的生長(zhǎng)有刺激作用,提高乳腺泌乳能力,增加產(chǎn)奶量[61-62];藻粉中的維生素E[63]、β-胡蘿卜素及其轉(zhuǎn)化形成的VA均可增強(qiáng)機(jī)體抗氧化能力[64],減緩氧化應(yīng)激,保證奶牛乳腺健康,提高產(chǎn)奶量和乳品質(zhì);藻粉中豐富的多糖、萜類化合物及維生素等活性物質(zhì)[65]可提高奶牛免疫機(jī)能,降低疾病發(fā)生的風(fēng)險(xiǎn),進(jìn)而保障乳產(chǎn)品的質(zhì)量和風(fēng)味。
4小結(jié)
鹽藻粉富含β-胡蘿卜素、多糖和蛋白等活性物質(zhì),在清除自由基和增強(qiáng)免疫力方面發(fā)揮重要作用,并可調(diào)控瘤胃發(fā)酵,提高營(yíng)養(yǎng)物質(zhì)利用率,改善奶牛泌乳性能。β-胡蘿卜素在動(dòng)物體內(nèi)可轉(zhuǎn)化為VA,因此添加鹽藻粉可彌補(bǔ)VA不足。綜上所述,鹽藻粉具有多種營(yíng)養(yǎng)生理功能,在奶牛生產(chǎn)中具有潛在應(yīng)用價(jià)值,但仍需更多的試驗(yàn)支持,其適宜添加量也需進(jìn)一步研究。

Abstract:The Dunaliella salina is a kind of unicellular eukaryote without cell wall,which is mainly used for extracting natural β-carotene and polysaccharides. The D. salina powder, which derives from the drying process of D. salina, plays a vital role in anti-oxidation, anti-virus, anti-tumor, and promotion of immune function. Currently, the applications of D. salina powder focused on human beings and rodents, but very few studies paid attention on dairy cows. We reviewed the regulation of rumen fermentation, oxidative stress, immune response, nutrient utilization and lactation performance by D. salina powder in dairy cows, aiming at providing references for dairy production.
