摘 要:? 桄榔子為桄榔(Arenga pinnata)的果實(shí),具有明顯的鎮(zhèn)痛活性。為研究桄榔子的化學(xué)成分,該文采用反復(fù)硅膠柱色譜、ODS柱色譜法并結(jié)合HPLC法進(jìn)行分離純化,并運(yùn)用 NMR 波譜技術(shù)鑒定化合物的結(jié)構(gòu),為植物資源的合理開(kāi)發(fā)和可持續(xù)利用提供科學(xué)依據(jù)。結(jié)果表明:從桄榔子70%乙醇提取物中分離得到 12 個(gè)黃酮類(lèi)化合物,分別鑒定為新落新婦苷(1)、甘草素(2)、染料木黃酮(3)、2, 4, 4′-三羥基查耳酮(4)、甘草苷(5)、染料木素-7-O-β-D-吡喃葡萄糖苷(6)、異甘草苷(7)、新異甘草苷(8)、大豆苷(9)、7-O-primverosylformononetin(10)、異甘草素-葡萄糖-芹糖苷(11)、毛蕊異黃酮苷(12)?;衔?-12均為首次從桄榔屬植物中分離得到。

Flavonoids from the Arenga pinnata

Abstract:? The Pinnata Fructus is the fruit of Arenga pinnata, which has obvious analgesic activity, but its chemical composition is rarely reported. In order to study the chemical compositions of the A. pinnata fruit, the compounds were separated and purified by repeated silica gel column chromatography and ODS column chromatography combined with HPLC, and the structures of the compounds were identified by using NMR spectroscopy. Twelve flavonoids were isolated from the 70% ethanol extraction of A. pinnata fruit, which were identified as neosmitilbin(1), glycyrrhizin(2), genistein(3), 2, 4, 4′-trihydroxychalcone(4), glycyrrhizin(5), genistein-7-O-β-D-glucopyranoside(6), isoglycyrrhizin(7), neo isoliquiritin(8), daidzin(9), 7-O-primverosylformononetin(10), icuraside(11), calycosin 7-O-β-glucoside(12). These twelve compounds were all isolated from A. pinnata fruit for the first time.

桄榔子為棕櫚科桄榔屬植物桄榔(Arenga pinnata)的干燥果實(shí),其功效為活血祛瘀、消食化積,用于婦女血脈阻滯之月經(jīng)不調(diào)、經(jīng)行不暢、小腹脹痛、產(chǎn)后瘀阻腹痛。始載于《開(kāi)寶本草》,謂其“主宿血”,《本草匯言》:“破宿食,積血。磨汁治婦人產(chǎn)后兒枕血瘕諸疼及心胃寒疼”。桄榔子在我國(guó)及東南亞國(guó)家有著悠久的應(yīng)用歷史,民間用于風(fēng)濕疼痛,止痛效果明顯。課題組前期通過(guò)藥效學(xué)研究證明桄榔子具有顯著的鎮(zhèn)痛抗炎活性(李鳳金等, 2019),對(duì)急性腹膜炎具有改善作用(李鳳金等, 2018),其作用機(jī)制與抑制炎癥因子的釋放、毛細(xì)血管通透性及抗氧化作用的降低、NF-κB和MAPK信號(hào)通路的激活的抑制、COX-2、IL-1β、TNF-α炎癥因子分泌較少有關(guān)(Li et al., 2019)。
目前,關(guān)于桄榔子化學(xué)成分文獻(xiàn)報(bào)道較少,課題組前期對(duì)桄榔子化學(xué)成分進(jìn)行系統(tǒng)研究,分離報(bào)道了18個(gè)化合物
(劉吉飛等, 2018;Liu et al., 2019)。本研究在此基礎(chǔ)上,重點(diǎn)對(duì)桄榔子中黃酮類(lèi)化合物進(jìn)行研究,分離得到12個(gè)黃酮類(lèi)化合物,分別鑒定為新落新婦苷(neosmitilbin)(1)、甘草素(glycyrrhizin)(2)、染料木黃酮(genistein)(3)、2,4,4′-三羥基查耳酮(2,4,4′-trihydroxychalcone)(4)、甘草苷(glycyrrhizin)(5)、染料木素-7-O-β-D-吡喃葡萄糖苷(genistein-7-O-β-D-glucopyranoside)(6)、異甘草苷(isoglycyrrhizin)(7)、新異甘草苷(neo isoliquiritin)(8)、大豆苷(daidzin)(9)、7-O-primverosylformononetin(10)、異甘草素-葡萄糖-芹糖苷(icuraside)(11)、毛蕊異黃酮苷(calycosin 7-O-β-glucoside)(12)。所有化合物均為首次從桄榔屬植物中分離得到,為我國(guó)廣西、廣東、福建,以及東南亞國(guó)家豐富的桄榔子資源開(kāi)發(fā)與利用奠定物質(zhì)基礎(chǔ)。
1 材料與儀器
桄榔子藥材于2017年9月采自廣西北海市,經(jīng)黑龍江省中醫(yī)藥科學(xué)院王偉明研究員鑒定為棕櫚科桄榔屬植物桄榔(Arenga pinnata)的果實(shí)。標(biāo)本保存于黑龍江省中醫(yī)藥科學(xué)院中藥研究所。
超導(dǎo)核磁共振儀(型號(hào):AV-400 型;廠家:瑞士 Bruker 公司);高分辨質(zhì)譜儀(型號(hào):Triple-TOFTM 5600+型;廠家:美國(guó) AB SCIEX 公司);高效液相色譜(型號(hào):515-2414 半制備型;廠家:美國(guó) Waters 公司);高效液相色譜柱(型號(hào):Cosmosil5C18 MS-Ⅱ制備型;規(guī)格:250 mm×20 mm,10 μm;廠家:日本Nacalai Tesque公司);高效液相色譜柱(型號(hào):Cosmosil 5 C18 MS-Ⅱ分析型;規(guī)格:250 mm×4.6 mm,5 μm;廠家:日本Nacalai Tesque公司);柱色譜硅膠(規(guī)格:80~120 目;廠家;青島海洋化工有限公司);薄層色譜硅膠G(青島海洋化工有限公司);反相柱色譜用填料(型號(hào):ODS-A-HG;規(guī)格:10 μm;廠家:日本YMC公司);甲醇(級(jí)別:色譜級(jí);廠家:德國(guó)默克公司);其余均為分析純。
2 提取與分離
取桄榔的鮮果約85 kg,切碎,以8倍量70%乙醇溶液室溫浸泡14 d,過(guò)濾,收集濾液,60 ℃減壓濃縮得浸膏約3 kg。浸膏按每100 g加水1 L 分散,用1∶1正丁醇萃取3次,并將萃取液80 ℃減壓濃縮至浸膏狀,得正丁醇浸膏約380 g。經(jīng)正相硅膠柱層析,用二氯甲烷-甲醇(2∶1→10∶1→5∶1→3∶1→2∶1→1∶1)6個(gè)梯度洗脫,TLC檢測(cè)合并為9個(gè)組分(Fr. A-I)。9個(gè)組分分別經(jīng)ODS純化,流動(dòng)相用甲醇-水(1∶9→2∶8→3∶7→4∶6→5∶5→6∶4→7∶3→1∶0),即(Fr. A1-10~I(xiàn)1-10)。Fr. F1再經(jīng)過(guò)制備型HPLC純化(C18柱, 55%甲醇-水, 流速5 mL·min-1 ),得到化合物7(6 mg);Fr. G5再經(jīng)過(guò)制備型HPLC純化(C18柱, 43%甲醇-水, 流速5 mL·min-1 ),得化合物2(8 mg)、4(8 mg);Fr. H4再經(jīng)過(guò)制備型HPLC純化(C18柱, 25%甲醇-水, 流速5 mL·min-1 ),得到化合物3(15 mg)、Fr. H2(C18柱, 35%甲醇-水, 流速5 mL·min-1 )得到化合物5(6 mg)、Fr. H3(C18柱, 45%甲醇-水, 流速5 mL·min-1 )得到化合物12(10 mg)、6(12 mg)、Fr. H5(C18柱, 55%甲醇-水, 流速5 mL·min-1 )得到化合物1(15 mg)、8(15 mg);Fr. I1再經(jīng)過(guò)制備型HPLC純化(C18柱, 15%甲醇-水, 流速5 mL·min-1 ),得到化合物9(5 mg)、Fr. I3(C18柱, 10%甲醇-水, 流速5 mL·min-1 )得化合物10(7 mg)、(C18柱, 60%甲醇-水, 流速5 mL·min-1 )得化合物11(8 mg)。
3 結(jié)構(gòu)鑒定

4 討論與結(jié)論
桄榔子作為一種具有鎮(zhèn)痛療效的民間藥,其藥效物質(zhì)基礎(chǔ)研究十分薄弱。棕櫚科藥用植物化學(xué)成分報(bào)道多集中于檳榔屬(Areca)植物檳榔(Areca catechu)的生物堿類(lèi)成分。而課題組通過(guò)系統(tǒng)分離研究發(fā)現(xiàn)桄榔子主要含有黃酮類(lèi)和酚酸類(lèi)成分,不含生物堿類(lèi)成分,說(shuō)明同科不同屬植物成分種類(lèi)差異較大。
前期研究發(fā)現(xiàn)桄榔子乙醇提取物及有效部位具有較強(qiáng)的鎮(zhèn)痛和抗炎作用,并申請(qǐng)了國(guó)家發(fā)明專(zhuān)利。該有效部位無(wú)中樞鎮(zhèn)痛作用,而外周鎮(zhèn)痛抗炎作用與一線(xiàn)非甾體抗類(lèi)風(fēng)濕性關(guān)節(jié)炎藥物洛索洛芬鈉相當(dāng)。非甾體抗炎藥作為一線(xiàn)抗風(fēng)濕藥,在發(fā)揮抗炎鎮(zhèn)痛作用的同時(shí),其嚴(yán)重的不良反應(yīng),如上消化道出血、肝腎不良反應(yīng)等,其中惡性心腦血管事件可能危及患者生命。本文分離獲得的12 個(gè)黃酮類(lèi)化合物是桄榔子有效部位的主要物質(zhì)基礎(chǔ),下一步應(yīng)通過(guò)活性組分的藥效學(xué)及鎮(zhèn)痛抗炎機(jī)理的深入研究,明確其藥效物質(zhì)基礎(chǔ)及作用靶點(diǎn)。為后續(xù)臨床應(yīng)用及質(zhì)量控制提供科學(xué)依據(jù),促進(jìn)桄榔子抗類(lèi)風(fēng)濕性關(guān)節(jié)炎等新藥研發(fā)及資源開(kāi)發(fā)利用。
