【摘 要】目的 通過體外及人體聯(lián)合測(cè)試方法,評(píng)價(jià)一款含泰束膠原蛋白肽護(hù)膚品的安全性及抗皺、緊致等方向的抗老化功效作用。方法 通過利用培養(yǎng)成纖維細(xì)胞,運(yùn)用ELISA方法檢測(cè)Ⅰ型膠原蛋白、彈性蛋白含量及第三方人體功效實(shí)驗(yàn)室(SGS)進(jìn)行人體功效檢測(cè),分別在0、2、4周采集分析皺紋、皮膚水分含量、經(jīng)皮水分流失值、彈性等數(shù)據(jù),綜合評(píng)估本產(chǎn)品的抗皺功效。結(jié)果 基于體外UVA刺激成纖維細(xì)胞模型試驗(yàn)得出,測(cè)試產(chǎn)品能顯著性提升Ⅰ型膠原蛋白、彈性蛋白含量(P<0.05);33名敏感肌受試者使用產(chǎn)品28 d后與使用產(chǎn)品前相比,皮膚角質(zhì)層水分含量、皮膚經(jīng)表皮失水值、皮膚彈性(R2值、R7值)、皮膚緊致F4、眼角處皺紋(長度、數(shù)量)均有極顯著性改善(P<0.001),皮膚彈性R5值與眼角處皺紋深度也得到改善(P<0.05)。結(jié)論 含泰束膠原蛋白肽精華對(duì)面部皮膚老化引起的皮膚干燥、粗糙和皺紋有一定的改善作用,且安全性良好。

Study on the Anti-aging Efficacy of A Skin Care Product Containing Taishu Collagen Peptides

【Abstract】Objective To evaluate the safety and anti-aging effect of a skin care product containing Taishu collagen peptide by in vitro and human joint test methods. Methods By using cultured fibroblasts, ELIASA method was used to detect the content of type Ⅰ collagen and elastin, and a third-party human efficacy laboratory (SGS) was used to detect human efficacy. The wrinkles, skin moisture content, transcutaneous water loss value, elasticity and other data were collected and analyzed at 0, 2 and 4 weeks to comprehensively evaluate the anti-wrinkle effect of the product. Results Based on the model test of fibroblasts stimulated by UVA in vitro, the test products could significantly increase the content of type Ⅰ collagen and elastin (P<0.05). After 28 days of using the product, the water content of skin stratum corneum, skin transepidermal water loss value, skin elasticity (R2 value, R7 value), skin tightening F4, and wrinkles (length, number) at the corner of the eye were significantly improved compared with those before using the product (P<0.001). The R5 value of skin elasticity and the depth of wrinkles at the corner of the eye were also improved(P<0.05). Conclusion Taishu collagen peptide essence on facial skin aging caused by dry skin, rough and wrinkles have a certain improvement, and with good safety.

皮膚老化是影響面部年輕化的重要原因,而紫外線照射又是外源性皮膚老化的關(guān)鍵因素,長期接觸紫外線的皮膚表現(xiàn)為粗糙、皺紋加深、失去彈性,伴有色素沉著[1]。隨著人們生活水平日益提高,越來越多的人們開始追求擁有健康、年輕的皮膚狀態(tài)。而泰束膠原蛋白肽是納米級(jí)純化學(xué)合成仿生人源Ⅰ型膠原蛋白,擁有Gly-Pro-Hyp膠原特征序列最多的肽段,富含約超過35%羥脯氨酸含量,比市面上大多數(shù)膠原蛋白中羥脯氨酸高13%[2],且分子量在2000 Da,可有效滲入皮膚角質(zhì)層[3],發(fā)揮抗衰功效。本研究通過研究泰束膠原蛋白肽對(duì)Ⅰ型膠原蛋白含量及彈性蛋白含量合成的影響,觀察該活性原料用于人體后的功效表現(xiàn),以期獲知基于泰束膠原蛋白肽的活性物在產(chǎn)品配方中體外和人體功效數(shù)據(jù),判斷其是否具有改善皮膚干燥、粗糙和皺紋的功效作用,現(xiàn)報(bào)道如下。
1.1 主要儀器與試劑 CO2培養(yǎng)箱(Thermo,150 L)、超凈工作臺(tái)(蘇州安泰,SW-CJ-1F)、酶標(biāo)儀(BioTek,Epoch)、倒置顯微鏡(Olympus,CKX41)、UVA照射儀(博溪生物)、Antera 3D(芬蘭Delfin)、Cutometer MPA580(德國CK)、Corneometer CM825(德國CK)、Tewameter TM Hex(德國CK);低糖DMEM培養(yǎng)基(博溪生物)、PBS(博士德)、新生牛血清(NBS,蘭州榮曄)、MTT(Sigma)、Elastin ELISA試劑盒(Abcam)、Collagen I alpha 1 ELISA試劑盒(Abcam)、TGFβ1(Peprotech)、覓光環(huán)游記芯肌多效修護(hù)緊致精華液(西安博鴻生物技術(shù)有限公司,陜G妝網(wǎng)備字2022000820,規(guī)格:30 ml)。
1.2 體外測(cè)試方法
1.2.1細(xì)胞毒性測(cè)試 按8×103個(gè)/孔的接種密度接種成纖維細(xì)胞至96孔板,培養(yǎng)箱(37 ℃,5% CO2)中孵育過夜。待96孔板中細(xì)胞鋪板率達(dá)到40%~60%時(shí)進(jìn)行給藥。溶劑對(duì)照組每孔加入200 μl 10% DMSO的培養(yǎng)液;調(diào)零組無細(xì)胞接種,僅加入200 μl細(xì)胞培養(yǎng)液;樣品組共設(shè)置8個(gè)濃度梯度,依次每孔加入200 μl 0.0781%,0.1563%,0.3125%,0.6250%,1.25%,2.5%,5%,10%(V/V)的泰束膠原蛋白肽精華液;以上三組均放置在培養(yǎng)箱中培養(yǎng)24 h,且每個(gè)樣品共設(shè)置3個(gè)重復(fù)孔。培養(yǎng)24 h后,棄掉上清,加入MTT工作液(0.5 mg/ml),37 ℃避光孵育4 h,孵育結(jié)束后,棄掉上清,每孔加150 μl DMSO,在490 nm處讀取OD值。利用細(xì)胞相對(duì)活力(%)=(樣品孔OD-調(diào)零孔OD)/(溶劑對(duì)照組OD-調(diào)零孔OD)×100%公式計(jì)算細(xì)胞相對(duì)活力。當(dāng)相對(duì)細(xì)胞活力≥80%,認(rèn)為受試物在該濃度下無細(xì)胞毒性。
1.2.2體外功效測(cè)試 按2.2×105個(gè)/孔的接種密度接種成纖維細(xì)胞至6孔板,培養(yǎng)箱(37 ℃,5% CO2)中孵育過夜;待6孔板中細(xì)胞鋪板率達(dá)到40%~60%時(shí),進(jìn)行分組給藥,每孔加樣2 ml,每組設(shè)3個(gè)復(fù)孔。給藥完成后將6孔板放置在培養(yǎng)箱(37 ℃,5% CO2)中繼續(xù)培養(yǎng)24 h。按照30 J/cm2輻照劑量對(duì)細(xì)胞進(jìn)行UVA輻照,繼續(xù)放入培養(yǎng)箱(37 ℃,5% CO2)中培養(yǎng)24 h后,收集細(xì)胞培養(yǎng)上清液于離心管中,根據(jù)Ⅰ型膠原蛋白、彈性蛋白ELISA試劑盒的操作說明書進(jìn)行檢測(cè)[4-6]。
1.3 人體測(cè)試方法 受試者選取上海地區(qū)自認(rèn)為敏感皮膚的31名亞洲女性和2名亞洲男性,年齡29~60歲;納入標(biāo)準(zhǔn):面部有輕度皮膚老化表現(xiàn)(干燥、細(xì)紋、彈性下降和皺紋等);自愿參加本試驗(yàn),簽署知情同意書,按要求使用產(chǎn)品并完成隨訪;近2個(gè)月內(nèi)沒有參加任何涉及皮膚的臨床試驗(yàn)。排除標(biāo)準(zhǔn):測(cè)試部位有皮膚疾病者;處于懷孕期、哺乳期、備孕期;經(jīng)研究者判斷為不適合參加試驗(yàn)者。按照每日早晚潔面后各使用1次,每次根據(jù)使用習(xí)慣量取適量產(chǎn)品,均勻涂抹于面部,輕輕按摩至吸收,連續(xù)使用28 d。使用Antera 3D(芬蘭Delfin)和Cutometer MPA580(德國CK)檢測(cè)使用前,使用后28 d隨機(jī)一側(cè)外眼角區(qū)域皺紋長度、皺紋平均深度、皺紋數(shù)量;皮膚彈性參數(shù)R2值、R5值、R7值和皮膚緊致參數(shù)F4值;并使用Corneometer CM825(德國CK)和Tewameter TM Hex(德國CK)檢測(cè)使用前、使用后14 d、28 d皮膚角質(zhì)層水分含量和經(jīng)皮水分流失值;測(cè)試時(shí),受試者清潔面部,并用無屑干面巾紙擦干皮膚,在溫度(21±2) ℃,濕度(50±10) %的檢測(cè)室中靜坐平衡30 min。各檢測(cè)指標(biāo)面部區(qū)域標(biāo)記點(diǎn)如下所示:
1.4 統(tǒng)計(jì)學(xué)方法 采用SPSS 28.0統(tǒng)計(jì)學(xué)軟件進(jìn)行數(shù)據(jù)分析,計(jì)量資料以(x-±s)表示,若測(cè)試數(shù)據(jù)為正態(tài)分布,則采用配對(duì)t檢驗(yàn)方法;若測(cè)試數(shù)據(jù)為非正態(tài)分布,則采用秩和檢驗(yàn);P<0.05表示差異有統(tǒng)計(jì)學(xué)意義,P<0.001表示統(tǒng)計(jì)學(xué)意義極顯著。
2.1 體外安全性測(cè)試結(jié)果 根據(jù)MTT和形態(tài)學(xué)結(jié)果,認(rèn)為樣品基于成纖維細(xì)胞,在1.25%的濃度范圍內(nèi)未表現(xiàn)出明顯的細(xì)胞毒性,見圖2、圖3。故使用該濃度進(jìn)行體外功效測(cè)試。
