      共軛聚合物熒光“開(kāi)關(guān)”的構(gòu)建及對(duì)S腺苷甲硫氨酸的分子識(shí)別與檢測(cè)

      1引言

      細(xì)胞內(nèi)的甲基化反應(yīng)存在通用甲基供體S腺苷甲硫氨酸(SAdenosylmethionine,AdoMet,SAM),SAM含有活性甲基,細(xì)胞內(nèi)幾乎所有用于甲基化修飾的甲基都來(lái)自SAM甲硫高能鍵。由于甲基化反應(yīng)的廣泛性,SAM是細(xì)胞內(nèi)參加反應(yīng)的重要性僅次于ATP的一種輔酶。細(xì)胞內(nèi)SAM濃度的微小改變,便會(huì)對(duì)細(xì)胞的生長(zhǎng)、分化和功能產(chǎn)生重大影響\[1\]。因此,實(shí)現(xiàn)快速、靈敏、準(zhǔn)確地檢測(cè)SAM具有重要意義。

      目前,已報(bào)道的測(cè)定SAM的方法有電化學(xué)法\[2\]、HPLC法\[3,4\]和分光光度法\[5\]。光度法依賴甲基轉(zhuǎn)移酶、S腺苷高半胱氨酸核苷酶和S核糖基高半胱氨酸酶的催化作用,將SAM分解為高半胱氨酸,再對(duì)高半胱氨酸顯色反應(yīng),操作比較繁瑣,準(zhǔn)確度也不理想。熒光光度法具有操作簡(jiǎn)單、靈敏度高、選擇性好等特點(diǎn),作為熒光傳感器的有小分子\[6,7\]、聚合物\[8,9\]、量子點(diǎn)\[10\]等。近年來(lái),基于熒光試劑和分析物質(zhì)之間競(jìng)爭(zhēng)作用使熒光試劑的熒光恢復(fù)得到較多關(guān)注\[11,12\]。水溶性熒光共軛聚合物\[13,14\]作為一類新型熒光傳感材料,具有摩爾吸光系數(shù)大、熒光量子產(chǎn)率高等優(yōu)點(diǎn),還具有“分子導(dǎo)線”的功能,能夠使熒光信號(hào)放大,常用于生物大分子和生物小分子的檢測(cè)\[15,16\]。

      本研究采用的水溶性陽(yáng)離子共軛聚合物聚3(1(2三乙胺乙?;┻哙?亞甲基)噻吩 (poly 3{\[1(2hydrazino2oxoethyl)piperidin4ylidene\]methyl}thiophene,PMTH),是具有優(yōu)良光電性能的熒光材料\[17,18\]。研究表明, Pb2+對(duì)PMTH熒光猝滅具有特異性,當(dāng)體系存在SAM時(shí),SAM與Pb2+的配位作用很強(qiáng),使PMTH從Pb2+PMTH配合物中被置換出來(lái),PMTH的熒光性能又恢復(fù),開(kāi)啟了PMTH的“開(kāi)關(guān)”熒光信號(hào)。本方法可以直接檢測(cè)SAM,不需要酶催化。

      3.4pH值的影響

      由于SAM結(jié)構(gòu)中有羥基、氨基和羧基,所以受體系pH值影響較大。實(shí)驗(yàn)表明,pH≤4.5時(shí),即在比較強(qiáng)酸性條件下,SAM與Pb2+很難形成配合物;pH>4.5時(shí),SAM與Pb2+可以形成配合物; pH>8.5時(shí),Cu2+有干擾;pH=4.5~9.0之間Pb2+與SAM以1∶1的比例形成配合物??疾炝薖b2+與PMTH反應(yīng)的pH值范圍,結(jié)果表明, pH=5.6~8.2時(shí)熒光強(qiáng)度達(dá)到最大,而且穩(wěn)定,根據(jù)體系中兩個(gè)反應(yīng)情況,選擇加入1.0 mL pH=7.2 TrisHCl緩沖溶液。

      3.5孵育時(shí)間和溫度對(duì)熒光恢復(fù)的影響

      當(dāng)所有的試劑都加入后,熒光強(qiáng)度恢復(fù)程度逐漸增大,在室溫下孵育10 min,熒光強(qiáng)度恢復(fù)程度達(dá)到最大且穩(wěn)定,且在5 h內(nèi)熒光強(qiáng)度相對(duì)穩(wěn)定,所以測(cè)定要在5 h內(nèi)完成。溫度升高, 孵育的時(shí)間縮短,但穩(wěn)定的時(shí)間也相對(duì)縮短,提高溫度不能提高體系的靈敏度,所以本實(shí)驗(yàn)采用在室溫條件下孵育10 min。

      4結(jié)論

      合成了一種水溶性共軛聚噻吩衍生物PMTH,通過(guò)對(duì)Pb2+與PMTH互相作用的熒光光譜的研究,考察了Pb2+對(duì)PMTH的熒光猝滅性能。在Pb2+PMTH的體系中加入SAM后,由于SAM與Pb2+的配位能力更強(qiáng), SAM可與Pb2+形成配合物,將Pb2+從PMTH鏈上置換出來(lái),使PMTH的熒光強(qiáng)度恢復(fù),恢復(fù)的熒

      光強(qiáng)度與SAM的濃度在一定范圍內(nèi)呈良好的線性關(guān)系。通過(guò)PMTH的熒光開(kāi)關(guān)信號(hào),成功建立了識(shí)別和檢測(cè)SAM的新方法。本方法具有高靈敏、高選擇性, 可簡(jiǎn)便快速、無(wú)標(biāo)記檢測(cè)SAM,并成功用于人血清、尿液及針劑樣品中SAM的檢測(cè)。
