摘要 [目的]研究樺褐孔菌的潛在應(yīng)用價(jià)值,并分析其菌絲體多糖的降糖作用。[方法]以生物量和粗多糖含量為指標(biāo),通過單因素試驗(yàn)與正交試驗(yàn)設(shè)計(jì)方法對其發(fā)酵條件進(jìn)行優(yōu)化。將樺褐孔菌菌絲多糖對糖尿病模型小鼠進(jìn)行灌胃,測定小鼠血糖變化。[結(jié)果]促進(jìn)該菌株發(fā)酵生長的最佳碳源、氮源分別為葡萄糖和牛肉膏;最佳培養(yǎng)基組成為葡萄糖30 g/L,蛋白胨4 g/L,牛肉膏8 g/L。搖床培養(yǎng)條件為pH 7.0,裝液量90 mL/250 mL,接種量15%,發(fā)酵時間12 d。與模型組相比,樺褐孔菌菌絲多糖能顯著降低糖尿病小鼠血糖。[結(jié)論]該研究可為工業(yè)化生產(chǎn)樺褐孔菌多糖及實(shí)際應(yīng)用提供參考。

Optimization of Fermentation Conditions of ?Inonotus obliquus ?and Research on Hypoglycemic ?in vitro

Abstract [Objective] To evaluate the potential application value of ?Inonotus obliquus ?and analyze the hypoglycemic effect of mycelium polysaccharide.[Method]Using biomass and crude polysaccharide yield as indicators,the fermentation conditions of ?Inonotus obliquus ?were optimized by single factor and orthogonal experiments.The diabetic model mice were intragastrically inoculated with ?Inonotus obliquus ?mycelium polysaccharide,and blood glucose changes in the mice were measured.[Result]The results showed that the most suitable carbon source and nitrogen source are glucose and beef extract,respectively.The optimum fermentation conditions were 30? g/L glucose,4? g/L peptone,8 g/L beef extract.Table concentrator culture conditions were pH 7.0,loaded liquid 90? mL/250? mL,inoculation amount 15% and fermentation time 12 d.Compared with the model group, Inonotus obliquus ?mycelium polysaccharide can significantly reduce blood glucose in diabetic mice.[Conclusion]The study can provide reference for the industrial production of ?Inonotus obliquus ?polysaccharide and its practical application.

樺褐孔菌 (Inonotus obliquus) 是一種木生白腐菌,因其具有抗氧化、抗癌、抗病毒和增強(qiáng)機(jī)體免疫等方面的作用[1],所以一直被當(dāng)作藥用真菌使用[2]。隨著對樺褐孔菌研究的不斷深入,已有研究者將樺褐孔菌用于惡性腫瘤、糖尿病和心血管疾病等重大疾病的預(yù)防和治療。由此可見,樺褐孔菌是一種極具開發(fā)價(jià)值的藥食兩用真菌[3-5]。樺褐孔菌野生品種生長周期長,需在活的樺樹上生長十余年才具有藥用作用[6-9],所以遠(yuǎn)不能滿足人們的需求。但是樺褐孔菌菌絲適應(yīng)性較強(qiáng),在普通的馬鈴薯葡萄糖瓊脂(Potato Dextrose Agar,PDA)培養(yǎng)基上即可生長[10-12]。發(fā)酵培養(yǎng)因其具有產(chǎn)量高、用時短、受周圍環(huán)境限制小、重復(fù)率高等優(yōu)點(diǎn),不失為培養(yǎng)樺褐孔菌的一種好選擇[13]。
