虎舌紅Ardisia mamillata Hance,又名紅毛氈、老虎舌、紅地氈、毛涼傘、金絲紅珠等,為紫金??谱辖鹋僦参?,原產(chǎn)我國(guó),分布于福建、廣西、廣東、云南、四川、貴州、江西等地[1],全株具有清熱利濕、平肝解郁、活血化淤的功效[2],是民間常用草藥。目前從虎舌紅植株中已經(jīng)分離得到三萜類、皂苷類、生物堿類、黃酮類、糖苷類、蒽醌類、酚醚類和甾體類等[3-6]。皂苷類主要為齊墩果烷型五環(huán)三萜衍生物,據(jù)報(bào)道,這類成分具有抗腫瘤、抗HIV 等活性[4]。課題組在前期研究[5-6]的基礎(chǔ)上,對(duì)虎舌紅根莖三萜皂苷類進(jìn)一步研究,從中分離得到7 個(gè)化合物,其中化合物4 ~7 為首次從該植物中分離得到,化合物5 為首次從該屬植物中分離得到。
IR Prestige-21 型傅立葉變換紅外光譜儀(美國(guó)Nicolet 公司);Bruker DRX-500 核磁共振儀(德國(guó)Bruker 公司);Thermo LCQ FLEET 型電噴霧質(zhì)譜儀(美國(guó)Thermo 公司);Agilent 1100 Series 液相色譜儀(美國(guó)Agilent 公司);YMC-Pack ODSAM 色譜柱 (4.6 mm×250 mm,5 μm); 凝膠SephadexLH-20(美國(guó)Pharmacia 公司);GF254薄層色譜和柱層析硅膠(青島海洋化工廠);乙腈為色譜純,其余試劑均為分析純。
植物樣品采自廣州農(nóng)業(yè)科學(xué)院白云實(shí)驗(yàn)基地,經(jīng)仲愷農(nóng)業(yè)工程學(xué)院植物教研室周俊輝教授鑒定為紫金??谱辖鹋僦参锘⑸嗉tArdisia mamillata Hance 的干燥根莖,標(biāo)本現(xiàn)存于本實(shí)驗(yàn)室。
干燥的虎舌紅根莖(7.5 kg) 粉碎,用80%乙醇加熱回流提取3 次,每次2 h,合并濾液,減壓濃縮,得乙醇浸膏1.05 kg,加水混懸,依次用石油醚、乙酸乙酯和正丁醇各萃取3 次,減壓濃縮各萃取液,得到乙酸乙酯部分96 g、 正丁醇部分412 g。正丁醇萃取物上D101 大孔吸附樹脂,依次用35%、75%、95%乙醇洗脫,收集75%乙醇洗脫部分,回收溶劑后得褐色浸膏21.6 g,將此浸膏進(jìn)行硅膠柱層析分離,氯仿-甲醇(100 ∶1 ~0 ∶100)梯度洗脫,合并相同流份,得到4 個(gè)組分(Fr.1 ~4)。Fr.1 濃縮后析出白色粉末,經(jīng)重結(jié)晶得化合物1(96 mg),經(jīng)制備HPLC 分離得化合物2(26 mg);Fr.2 經(jīng)ODS 柱層析和重結(jié)晶得化合物3(59 mg),經(jīng)制備HPLC 分離得化合物4 (57 mg);Fr.3 經(jīng)反復(fù)硅膠柱層析、凝膠層析和制備HPLC分離得到化合物5(29 mg);Fr.4 經(jīng)Sephadex LH-20 反復(fù)柱層析純化,再用制備HPLC 分離純化得到化合物6(74 mg)、7(29 mg)。

結(jié)果表明,從虎舌紅分離得到7 個(gè)五環(huán)三萜類化合物,均為具有3 ~5 個(gè)糖的齊墩果烷型三萜皂苷?;衔? ~2 的苷元結(jié)構(gòu)為13,28-環(huán)氧-30-醛基(A 型);化合物3 ~4 的苷元結(jié)構(gòu)為13,28-環(huán)氧-16-羰基(B 型);化合物5 的苷元結(jié)構(gòu)為12-烯-30-羰基(C 型);化合物6 ~7 的苷元結(jié)構(gòu)為13,28-環(huán)氧醚 (D 型)、13,28-環(huán)氧醚-30-羰基 (E型),見圖1。

化合物1 對(duì)人卵巢癌細(xì)胞A2780 顯示出較強(qiáng)的細(xì)胞毒性[16]; 化合物2 對(duì)腫瘤細(xì)胞MCF-7、HepG2 和人紅白血病K562、人胃癌KATO-Ⅲ、人卵巢癌OVCA2780 細(xì)胞均顯示出較強(qiáng)的抑制作用[17-18];化合物4、6 ~7 對(duì)人肝癌細(xì)胞HepG2 均有較強(qiáng)的抑制細(xì)胞增殖的活性[10,13]??梢?,三萜皂苷是虎舌紅抗腫瘤活性的主要有效成分,以期本研究結(jié)果為進(jìn)一步闡明虎舌紅抗腫瘤作用的藥效物質(zhì)提供科學(xué)依據(jù)。
